Mutiple myeloma resistant to melphalan (NSC-8806) treated with cyclophosphamide (NSC-26271), prednisone (NSC-10023), and chloroquine (NSC-187208)

A prospective randomized study of the effects of two different therapeutic regimens was conducted in 41 patients with multiple myeloma resistant to therapy with melphalan. The results of treatment with cyclophosphamide plus prednisone were compared with those of cyclophosphamide, prednisone, and chloroquine. Chloroquine, which inhibits the repair process of DNA in animals, has reportedly produced responses to alkylating agents to which the animal had been resistant. No significant differences in response to the doses used with either regimen could be found. Toxocity consisted mainly of leukopenia and thrombocytopenia.

This study was a prospective randomized clinical trial conducted in 41 patients with multiple myeloma that was resistant to treatment with melphalan. The study aimed to compare the effectiveness of two different therapeutic regimens, namely, cyclophosphamide plus prednisone versus cyclophosphamide, prednisone, and chloroquine.

Chloroquine is a drug that inhibits the repair process of DNA in animals and has been reported to produce responses to alkylating agents in animals that were previously resistant to them. However, the use of chloroquine in this study did not result in any significant differences in response compared to the group that did not receive chloroquine.

Toxicity of both regimens was mainly related to low white blood cell count (leukopenia) and low platelet count (thrombocytopenia). No significant difference in toxicity was observed between the two groups.

It is important to note that this study had a small sample size, and further research with larger sample sizes may be necessary to confirm these findings.
