Procyanidin B1 is a polyphenolic flavonoid isolated from fruits and fruit juices, which inhibits LPS-induced TNF-α production, and expression of MD-2, TRAF6, NF-κB mRNA, phosphorylated p38 MAPK and NF-κB protein in THP1 cells.[1]

Method

THP1 cells are treated for 18 h with 1 µg/mL LPS in the presence or absence of 100 µg/mL procyanidin B1. Total RNA is extracted from THP1 cells using the RNAiso Plus kit.  Isolated total RNA is  reverse transcribed using an RT master mix kit for 15 min at 37℃ and 5 s at 85℃. cDNAs are amplified using a 7500 Real-Time PCR System with sequence-specific primers for MD-2, TLR4, TRAF6, and NF-kB designed and synthesized by Takara Biotechnology.

Procyanidin B1 selectively inhibits the Kv10.1 channel in a concentration-dependent manner, which significantly suppress the growth of the tumor (HepG2) with no side effect on the normal metabolismof the mice.[2]
