Description

Procyanidin B1, a natural polyphenolic compound that has antioxidant properties, inhibits Kv10.1 channel and suppresses the evolution of hepatoma.

In vitro

Procyanidin B1 is a polyphenolic flavonoid isolated from fruits and fruit juices, which inhibits LPS-induced TNF-α production, and expression of MD-2, TRAF6, NF-κB mRNA, phosphorylated p38 MAPK and NF-κB protein in THP1 cells.^[1]

In vivo

Procyanidin B1 selectively inhibits the Kv10.1 channel in a concentration-dependent manner, which significantly suppress the growth of the tumor (HepG2) with no side effect on the normal metabolismof the mice.^[2]

Cell Assay:^{^[1]}	Cell lines THP1 cells Concentrations 100 µg/mL Incubation Time 18 h Method THP1 cells are treated for 18 h with 1 µg/mL LPS in the presence or absence of 100 µg/mL procyanidin B1. Total RNA is extracted from THP1 cells using the RNAiso Plus kit. Isolated total RNA is reverse transcribed using an RT master mix kit for 15 min at 37℃ and 5 s at 85℃. cDNAs are amplified using a 7500 Real-Time PCR System with sequence-specific primers for MD-2, TLR4, TRAF6, and NF-kB designed and synthesized by Takara Biotechnology.

Method
THP1 cells are treated for 18 h with 1 µg/mL LPS in the presence or absence of 100 µg/mL procyanidin B1. Total RNA is extracted from THP1 cells using the RNAiso Plus kit.
