G6PDi-1 effectively inhibits G6PD. Across a range of cultured cells, G6PDi-1 depletes NADPH most strongly in lymphocytes. In T cells but not macrophages, G6PDi-1 markedly decreases inflammatory cytokine production. In neutrophils, it suppresses respiratory burst.[1]

Method

To analyze intracellular cytokines, active T cells are re-stimulated with PMA (20 ng/ml) and ionomycin (1 ug/ml) in the presence of GolgiStop and increasing concentrations of G6PDi-1. After a 6h incubation period, cells are collected, washed with PBS and stained with the viability dye Live/Dead Aqua.
For measuring intracellular ROS levels, cells are incubated for 2 h in the presence of increasing concentrations of G6PDi-1 and then stained for ROS and viability using CellROX Green Flow Cytometry Assay Kit following manufacturer instructions.
