Hydroxyflutamide is an androgen receptor (AR) antagonist, which enhances the effects of irradiation in AR-positive cells cultured in the presence of androgens.[2]

Method

Cells (2×105/dish) seeded in 35-mm tissue culture dishes and incubated in the presence or absence of DHT, HF, and/or inhibitors of DNA repair for 48 hours are irradiated using a Gammacell 40 Cs-137 irradiator (0.6 Gy/min). The irradiated cells are then used for MTT and clonogenic assays.

In a mouse xenograft model, low-dose flutamide are found to enhance the inhibitory effects of irradiation, and its tumor size is similar to that of AR knockdown line with radiation alone.[2]
