Rebaudioside A potentiates glucose-stimulated insulin secretion on MIN6 cells. It does not shange intracellular cAMP level of isolated mouse islets. Rebaudioside A modulates the effect of glucose on insulin secretion via indirect inhibition of the KATP channels, thereby allowing β cells to depolarize and generate action potentials, events that are important for glucose-stimulated insulin secretion[1].

Method

After 30 min of preincubation with Krebs-Ringer bicarbonate buffer without glucose, cells are exposed to the indicated conditions for 60 min; the solution is then collected and subjected to radioimmunoassay for the detection of insulin levels.
