JNJ0966 is a highly selective compound that inhibits activation of MMP-9 zymogen and subsequent generation of catalytically active enzyme. It has no effect on MMP-1, MMP-2, MMP-3, MMP-9, or MMP-14 catalytic activity and did not inhibit activation of the highly related MMP-2 zymogen.

Biological Activity

Description JNJ0966 is a highly selective compound that inhibits activation of MMP-9 zymogen and subsequent generation of catalytically active enzyme. It has no effect on MMP-1, MMP-2, MMP-3, MMP-9, or MMP-14 catalytic activity and did not inhibit activation of the highly related MMP-2 zymogen.
In vitro
In vitro JNJ0966 inhibits the conversion of proMMP-9 to active MMP-9 but is not a direct inhibitor of MMP-9 or MMP-3 enzymatic activity. JNJ0966 inhibits proMMP-9 maturation[1].
In Vivo
In vivo Oral administration of JNJ0966 significantly diminishes the clinical disease score observed in the mouse EAE model[1].
