CU-CPT8m, recognizes a pocket on the interface of two copies of TLR8 and prevents them from undergoing conformational changes that are necessary for the proinflammatory signaling.[2]

Method

HEK-Blue TLR8 cells were seeded in tissue culture treated 96-well plates with a density of 7.5 × 104 cells/well in DMEM media supplemented with 10% FBS (phosphatases deactivated with heat). These cells were then treated with R848 (1 μg/mL) along with various concentrations of appropriate compounds. Plates are then incubated in humidified incubators containing 5% CO2 at 37 °C for 24 h. Supernatant was removed, and 100 µL of 1:10 diluted WST-1 (Roche) was then added to each well. The plate was then incubated at 37 ℃ till color change (30 min-1 h).
