Biological Activity

Description Methyl salicylate (Betula oil, Gaultheria oil, Methyl 2-hydroxybenzoate, Natural wintergreen oil) is an organic ester naturally produced by many species of plants, particularly wintergreens. It is an agonist for TRPA1 and TRPV1.
Targets
TRPA1 [1]
(Cell-free assay)
TRPV1 [1]
(Cell-free assay)
In vitro
In vitro In human embryonic kidney 293 cells expressing human TRPV1 (hTRPV1), Methyl salicylate evokes increases of [Ca2+]i, which declines regardless of its continuous presence, indicative of marked desensitization. Methyl salicylate promotes Ca2+ influx through the activation of TRPV1 channels and reversibly inhibits hTRPV1 activation by polymodal stimuli such as capsaicin, protons, heat, anandamide, and 2-aminoethoxydiphenyl borate. Methyl salicylate is an effective agonist for recombinant human and porcine TRPV1[1].
Cell Research Cell lines HEK 293 cells
Concentrations 3 mM
Incubation Time --
Method Fura-2-loaded hTRPV1-expressing HEK 293 cells are first stimulated with capsaicin (30 nM) and subsequently with Methyl salicylate (3 mM).
In Vivo
In vivo Methyl salicylate significantly suppresses nocifensive behavior induced by intraplantar capsaicin in rats[1].
Animal Research Animal Models Male Wistar rats (8 weeks old)
Dosages 0.5 mg
Administration injected into the same hind paw

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL ddH2O, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such
as vortex, ultrasound or hot water bath can be used to aid dissolving.
