In vitro
In vitro Abietic acid has anti-inflammatory effects by inhibiting expression of genes involved in inflammation such as those for tumor necrosis factor-a and cyclooxygenase 2 in lipopolysaccharide-induced macrophages. Abietic acid regulates PPARγ target gene expression in RAW 264.7 macrophages and 3T3L adipocytes. It can be used for regulating lipid metabolism and atherosclelosis[2]. Abietic acid is a PPARγ ligand and that its anti-in£ammatory effect is partly due to the activation of PPARγ in stimulated macrophages. In lipopolysaccharide (LPS)-stimulated macrophages, abietic acid suppresses production of prostaglandin E2 (PGE2) in vitro and in vivo[1].

Method

pM-hPPARγ, p4xUASg-tk-luc, pCMX-CBP and pRL-CMV are transfected into CV1 cells and then, 24 h after the transfection, the cells are treated with abietic acid at various concentrations for 24 h.

In vivo Abietic acid has an antiobesity effect in mice mediated by the regulation of adipogenesis[2].
