Description

SMIFH2 is a specific inhibitor of formin that inhibits formin-driven actin polymerization in vitro with IC50s ranging from 5 to 15 μM for different formins.

SMIFH2 inhibits stress fiber contractility in living and permeabilized REF52 cells. SMIFH2 inhibits centripetal movement of myosin 2 filaments in living and permeabilized HFF cells. SMIFH2 inhibits activity of myosin ATPase and the ability to translocate actin filaments in the gliding actin.[1]

Method

Cells are seeded at density of 5×104 cells/ml on the hydrophobic uncoated μ-dish 35 mm with fibronectin micro-patterns and incubated 3-8 h prior to the experiment. Cells are permeabilized with extraction buffer A supplemented with 0.1% Triton X-100 and 4% PEG MW35000 for 10 min at room temperature, followed by three washes with extraction buffer A. Cytoskeleton contractility assay was carried out at 37°C using buffer A supplemented with 2 mM ATP with or without SMIFH2.

SMIFH2 induces tail curvature and peripheral cell death, craniofacial malformations, pericardial edema in developing zebrafish embryos. SMIFH2 impairs vascular flow and impacts heart rates in developing zebrafish embryos.[2]
