Description Dantrolene sodium(F 440) acts as a postsynaptic muscle relaxant by inhibiting Ca2+ ions release from sarcoplasmic reticulum stores by antagonizing ryanodine receptors.

In vitro

Treatment with a mixture of Thaps. and dantrolene during optogenetic stimulation (OS) of Schwann cells (SCs) significantly decreases the percentage of MBP-expressing SCs and, importantly, reduces the percentage of MBP-expressing SCs. Mixture of Thaps. and dantrolene decreases drastically the ΔF/F value in SCs.[1]

Method

Thaps. + Dantrolene (600 nM, a ryanodine receptor antagonist) for 4 h prior to examining the intracellular Ca2+ levels of SCs. Representative inverted images of BABTA-1 AM fluorescence signals showing the intracellular Ca2+ on SCs.

In vivo

Dantrolene administration decreases plasma GLuc-SERCaMP activity and endogenous SERCaMP activity when compared to vehicle during ad lib high fat diet. Rats administered dantrolene gains less weight compared to vehicle treated rats, and have less caloric intake.
