Biological Activity

Description Alamandine is a component of the renin-angiotensin system, which regulates blood pressure.
In vitro
In vitro

Alamandine decreases leptin secretion and expression in isolated adipocytes mediated by Gq, c-Src, p38 mitogen-activated protein and IκB activation, also induces nitric oxide expression via inducible nitric oxidase synthase and plasminogen activator inhibitor 1 expression in isolated adipocytes.[2]

Cell Research Cell lines adipocytes isolated from rats, 3T3L-1 adipocytes
Concentrations 0.01, 0.1, 1, 10, 100, 1000 nM
Incubation Time 24 h
Method

Isolated adipocytes are incubated with alamandine for 24 h. Secreted leptin levels are determined using a leptin immunoassay kit according to the manufacturer’s protocol.

In Vivo
In vivo

Moderately concentrated alamandine reduces blood leptin levels in vivo.[2]

Animal Research Animal Models Wistar rats (10-week-old)
Dosages 576, 57.6, 5.76, or 0.576 µg/kg
Administration i.p.

Method for preparing DMSO master liquid: mg drug pre-dissolved in μL DMSO ( Master liquid concentration mg/mL, Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug. )

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL ddH2O, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such
as vortex, ultrasound or hot water bath can be used to aid dissolving.
