Biological Activity

Description T3 (Triiodothyronine) is a thyroid hormone that affects various physiological process in the body, including growth and development, metabolism, body temperature, and heart rate.This product is soluble but easy to precipitate after freezing in DMSO. 0.1 M NaOH is recommended as a stock solution.

Triiodothyronine is one ingredient supplemented in the medium for inducing brown adipocytes differentiation.[1]

  • Method

    Brown preadipocytes were isolated from perirenal adipose tissues of Chuanzhong black goats (1 day after birth). The cells were cultured in DMEM/F12 containing 10% fetal bovine serum and 2% penicillin/streptomycin and placed in an incubator (5% CO2, 37°C). Until 100% confluence, DMEM/F12 was supplemented with 2% FBS containing 5 μg/ml of insulin, 0.5 mM IBMX, and 2 nM triiodothyronine to induce brown adipocytes differentiation. Two days later, the induced medium was changed to a maintenance medium (DMEM/F12 supplemented with 2% FBS, 5 μg/ml of insulin, and 2 nM triiodothyronine). Cells were harvested 8 days after differentiation. L-carnitine was added to the medium throughout the differentiation process and supplemented at 600 μΜ. SiAMPKα (GAAGAGAAATTCAGAACCT)15 were transfected by Lipofectamine 3000. Lipid droplets were stained using Oil Red O solution. After staining, 1 ml isopropanol was used to dissolve the oil red O dye, and the absorbance at 510 nm was detected.
