Biological Activity

Description Tolmetin(MCN2559) is a non-steroidal anti-inflammatory drug of the arylalkanoic acids. Tolmetin sodium dihydrate is a non-steroidal antiinflammatory agent, and acts as a non-selective COX inhibitor.
In vitro

Tolmetin is a nonsteroidal antiinflammatory drug (NSAID) which is useful in the treatment of rheumatoid arthritis, osteoarthritis, and gout. Tolmetin induces prostaglandin synthesis inhibition in vitro with IC50 of 3 mg/mL. [1]

In vivo

Tolmetin (100 mg/kg, po) produces a dose-dependent antinociceptive effect in rats received an intra-articular injection of uric acid in the right hindlimb, which is not directly related to blood concentration. [1] Tolmetin (100 mg/kg i.p.) is metabolized into tolmetin acyl-CoA (Tol-CoA), the acyl-CoA-dependent metabolites tolmetin-taurine conjugate (Tol-Tau) and tolmetin-acyl carnitine ester (Tol-Car) in liver homogenates of rats. [2] Tolmetin sodium (1 mg/mL in 5-15 mL) is consistently efficacious in reducing postoperative adhesion formation in uterine horn and peritoneal side wall of rabbits. Tolmetin sodium (1 mg/mL in 3 mL) is consistently efficacious in reducing postoperative adhesion formation in uterine horn and peritoneal side wall of rats. [3] Tolmetin (0.5-2.0 mg/mL) combined with hyaluronic acid is efficacious in reducing adhesion formation in a standardized rabbit model. [4] Tolmetin sodium (100 mg/kg, i.p.) transiently decreases the phagocytic capability of peritoneal leukocytes in normal rats. Tolmetin (100 mg/kg, i.p.) significantly elevates superoxide anion release at days 3 and 5, phagocytosis at days 7 and 14 and tumoricidal activity at day 3 in normal rats. [5]
