Biological Activity

Description Sodium metatungstate (SMT) solution is an inorganic heavy liquid which is widely used in density fractionation. Sodium metatungstate (Sodium polyoxotungstates) is a potent and selective inhibitor of ecto-nucleoside triphosphate diphosphohydrolase (NTPDases, CD39) with Ki of 0.140 μM, 0.910 μM and 0.563 μM for NTPDase1, NTPDase2 and NTPDase3, respectively.

In vitro

Sodium metatungstate, a inhibitor of CD39, can partially reverse the suppression of CD8[+H] T cells induced by the presence of peritoneal macrophage.[2]

  • Method

    Peritoneal lavage cells are plated in complete RPMI media on a 10 cm dish. Celexocib (10 μM), anti-TGF-b (clone 1D11, 10 μg/ml), or sodium metatungstate (50 μM) are plated onto respective wells of a 96 well round bottom plate. After at least five rounds of gentle aspiration and washing of the 10 cm plate containing the peritoneal lavage cells, adherent peritoneal macrophages are removed with a cell scraper, collected, and counted. 105 peritoneal macrophages are plated per indicated well of the 96 well plates. CD8+ T cells are purified from ACK-lysed splenocytes from C57BL/6 mice utilizing the Mouse CD8a+ T Cell Isolation Kit and then 105 purified CD8+ T cells are plated onto in T cell medium with 50 IU/ml murine IL-2. Certain wells are incubated with 105 anti-mouse CD3/CD28 Dynabeads for T cell activation. Flow cytometry is performed after 48 hours of culture.
