In vivo (Add solvents to the product individually and in order)
Clear solution
5%DMSO 40%PEG300 5%Tween80 50%ddH2O
2.5mg/ml Taking the 1 mL working solution as an example, add 50 μL of 50 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.
Clear solution
5% DMSO 95% Corn oil
2.5mg/ml Taking the 1 mL working solution as an example, add 50 μL of 50 mg/ml clear DMSO stock solution to 950 μL of corn oil and mix evenly. The mixed solution should be used immediately for optimal results.
* <1 mg/ml means slightly soluble or insoluble.
* Please note that Selleck tests the solubility of all compounds in-house, and the actual solubility may differ slightly from published values. This is normal and is due to slight batch-to-batch variations.
* Room temperature shipping (Stability testing shows this product can be shipped without any cooling measures.)

Biological Activity

Description Rimiducid (AP1903) is a chemical protein dimerizer and binds tightly to the target FKBP fusion protein while binding minimally to the abundant natural FKBP. Rimiducid elicites potent and dose-dependent apoptosis of cells in culture expressing dimerizer-dependent Fas constructs, with an EC50 of ≈0.1 nM.
Targets
FKBP fusion protein [1]
In vitro

Rimiducid (AP1903) induces effective and dose-dependent apoptosis in the culture expressing dimer-dependent Fas constructors, indicating that Rimiducid causes the activation of Fas signaling without interference from endogenous FKBP.<a class="sref" href="#s_ref">[2]</a>

In vivo

Rimiducid (AP1903) elicits a dose-dependent decrease in serum hGH levels, with a half-maximal effective dose of 0.4±0.1 mg/kg, which means AP1903 is functional of activating of Fas signaling in vivo.<a class="sref" href="#s_ref">[2]</a>

Protocol (from reference)

Cell Assay:

<a class="sref" href="#s_ref">[2]</a>

  • Cell lines

    HT1080 cells

  • Concentrations

    0.01-1000 nM

  • Incubation Time

    Overnight

  • Method

HT1080 cells stably transduced with retrovirus pSR a-myr-2FKBP-Fas-E (open circles) or pSR a-myr-2 (F36V-FKBP)- Fas-E (closed circles) were treated overnight with AP1903, and viability then was measured by Alamar Blue assay.

Animal Study:

<a class="sref" href="#s_ref">[2]</a>

  • Animal Models

Nude mice implanted with hGH-secreting HTFasGH-3 cells

  • Dosages

    2 ml/kg

  • Administration

    i.v.
