Biological Activity

Description Oleic acid (Oleate, 9-Octadecenoic Acid, Elaidoic acid) is a fatty acid that occurs naturally in various animal and vegetable fats and oils. It is used commercially in the preparation of oleates and lotions, and as a pharmaceutical solvent. Oleic acid is an abundant monounsaturated fatty acid. Oleic acid is a Na+/K+ ATPase activator.Oleic Acid can be used to induce animal models of Renal Failure.
In vitro Oleic acid treatment induces a marked increase in the proliferation of 786-O cells in a dose-dependent manner. It delays 786-O cell apoptosis. In 786‑O cells, ILK is upregulated by oleic acid treatment. Oleic acid activates G protein-coupled receptors and phosphorylates ERK1/2 to induce cancer cell proliferation in breast cancer[1]. Oleic acid(as low as 5 M) inhibits the stearic acid-induced increase in intercellular adhesion molecule-1 (ICAM-1) expression. Oleic acid is able to diminish endothelial cell activation, irrespective of the stimulus utilized, by suppressing the upregulation of adhesion molecules. It can protect Chinese hamster ovary cells from palmitate-induced lipotoxicity by promoting triglyceride synthesis within the cells[2].
In vivo Oleic acid inhibits in vivo Na/K-ATPase activity in the lungs and induced lung injury. 10 μM oleic acid induces lung edema and inflammation with cell influx, lipid body formation, and leukotriene B4 (LTB4) and prostaglandin E2 (PGE2) production[3]. Oleic acid ameliorates amyloidosis in mouse models of Alzheimer's disease[4].

  • Method

    Cells (2x103 cells/well) are seeded in 96-well microtitre plates and incubated for 24 h in 100 µl culture medium. Cells in the experimental group were then treated with 0.05, 0.1 or 0.2 mmol/l oleic acid for 48 h. MTT [20 µl (5 g/l)] is added to the cells which were then cultivated for a further 4 h. Following the removal of the supernatant fluid, 150 µl/well DMSO is added to the cells which are agitated for 15 min. Absorbance is measured at 490 nm by an ELISA reader.
