Description

NSC45586 is an inhibitor of PHLPP (Pleckstrin homology domain and leucine-rich repeat protein phosphatase), which is selective for PHLPP1 and PHLPP2.

PH domain and leucine rich repeat phosphatase (Phlpp) inhibitor NSC45586 increases Pth1r mRNA and protein expression in ATDC5 cells and IMCs within 24 hours. ^[2]

Cell Assay:^{^[2]}	Cell lines ATDC5 cells, Primary immature chondrocytes (IMCs) Concentrations 25 μM Incubation Time 24 h Method ATDC5 cells were seeded at a density of 5 x 105 cells/well in a six-well plate and incubated overnight to allow for cell adhesion in DMEM supplemented with 10% FBS and 100 units/mL penicillin, 100 μg/mL streptomycin, and 0.25 μg/mL Amphotericin B (1% antibiotic/antimycotic). Phlppi including NSC45586 (25 μM) treatments were added. Primary chondrocytes were plated and allowed to adhere overnight as described above. Actinomycin D (5μM) was added for six hours and then replaced with medium containing Phlppi including NSC45586 or vehicle for 24 hours. Cells were collected for RNA or protein extraction.
