Biological Activity

Description LC3-mHTT-IN-AN1 is a mHTT-LC3 linker compound that interacts with both mutant huntingtin protein (mHTT) and LC3B. LC3-mHTT-IN-AN1 reduces the levels of mHTT in an allele-selective manner in cultured Huntington disease (HD) mouse neurons.

    LC3-mHTT-IN-AN1 was diluted in culture medium to 10×concentrations and added to the plated cells: for primary cultured neurons and iPS-cell-derived neurons, LC3-mHTT-IN-AN1 was added 5 days after plating; The cells were then collected 2 days later for measurement of HTT levels. For detection of HTT-LC3 colocalization, the cells were fixed 4 h after compound treatment. For caspase-3 activation detection, the cells were stressed (BDNF removal for iPS-cell-derived neurons) 1 day after compound treatment, and tested at the indicated time points.
