Solubility (25°C)* In vitro DMSO 20 mg/mL (61.85 mM)
Water Insoluble
Ethanol Insoluble
In vivo (Add solvents to the product individually and in order)
Clear solution
5%DMSO 40%PEG300 5%Tween80 50%ddH2O
8.0mg/ml Taking the 1 mL working solution as an example, add 50 μL of 160 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.
* <1 mg/ml means slightly soluble or insoluble.
* Please note that Selleck tests the solubility of all compounds in-house, and the actual solubility may differ slightly from published values. This is normal and is due to slight batch-to-batch variations.
* Room temperature shipping (Stability testing shows this product can be shipped without any cooling measures.)

Biological Activity

Description iFSP1 is a potent, selective and glutathione-independent inhibitor of ferroptosis suppressor protein 1 (FSP1/AIFM2) with EC50 of 103 nM. iFSP1 selectively induces ferroptosis in GPX4-knockout Pfa1 and HT1080 cells that overexpresses FSP1.Solutions are unstable and should be fresh-prepared.
Targets
FSP1/AIFM2 [1]
(Cell-free assay)
103 nM(EC50)
In vitro

iFSP1 selectively induces ferroptosis in GPX4-knockout Pfa1 and HT1080 cells that overexpressed FSP1. FSP1 is expressed in most tumour cell lines, and iFSP1 treatment robustly sensitizes these cells to RSL3-induced ferroptosis.[1]

Protocol (from reference)

Cell Assay:

[1]

  • Cell lines

    human cancer cell lines (NCl-H1437, U-373, MDA-MB-436, SW620, MDA-MB-435S, A549, etc.)

  • Concentrations

    0.01 μM - 10 μM, 3 μM

  • Incubation Time

    24 h

  • Method

Cells are seeded on 96-well plates (2,500 cells per well) and treated with iFSP-1 after plating. Cell viability is assessed 24 h (unless stated otherwise) after treatment using AquaBluer as an indicator of viable cells according to the manufacturer's recommendations.
