Biological Activity

Description Glucosamine sulfate (D-Glucosaminesulfate, D-Glucosamine sulphate) is a dietary supplement commonly used in the treatment of osteoarthritis. It is a type of glycosaminoglycan, which is a naturally occurring major component of joint cartilage and joint fluid.
In vitro Glucosamine sulfate exerts a pronounced suppressive effect on MMPs in osteosarcoma cell lines in vitro[1]. Glucosamine suppresses the activation of T-lymphoblasts and dendritic cells in vitro as well as allogeneic mixed leukocyte reactivity in a dose-dependent manner. Glucosamine has been shown to inhibit nucleic acid and protein biosynthesis and irreversible damage to organelles in tumor cells, but not in normal cells[3].
In vivo Glucosamine sulfate attenuates DSS-induced colitis symptoms, possibly by suppressing the NF-κB-mediated TNF-α and IL-1β production and neutrophil activation that contribute to intestinal barrier disruption in colitic mice[2]. Glucosamine administration prolongs allogeneic cardiac allograft survival in vivo[3].

Protocol (from reference)

Cell Assay:[1]
  • Cell lines

    MG-63 and SaOS-2 cells

  • Concentrations

    0, 10, 50 and 100 μg/ml

  • Incubation Time

    48 h

  • Method

Cells are exposed to 0, 10, 50 and 100 μg/ml glucosamine sulfate for 48 h and treatment toxicity is determined through measurement of cell viability and proliferation. Relative gene expression of matrix metalloproteinase (MMP)-2, -3 and -9 is quantified by real-time polymerase chain reaction. Protein levels of MMP-2 and -9 are assessed by ELISA.

Animal Study:[2]
  • Animal Models

    C57BL/6 mice

  • Dosages

    0, 0.05, and 0.10% (w/w)

  • Administration

    oral
