Taking the 1 mL working solution as an example, add 50 μL of 100 mg/ml clarified H2O stock solution to 400 μL of PEG 300, mix evenly to clarify; add 50 μL of Tween 80 to the above system, mix evenly to clarify; Then continue to add 500 μL ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Description

Colivelin (CLN) is a brain-penetrant neuroprotective peptide with potent long-term capacity against Aβ deposition, neuronal apoptosis, and synaptic plasticity deficits in neurodegenerative disease. Colivelin is an activator of STAT3.

In vitro

Colivelin, a brain penetrant neuroprotective peptide, significantly increases the p-STAT3 protein levels in BV-2 cells.^[2]

In vivo

Colivelin protects against ischemic brain injury and improves neurological outcomes, by elevating anti-apoptotic genes up-regulation and activates JAK/STAT3 signaling afetr ischemic stroke.^[1]

Cell Assay:^{^[2]}	Cell lines BV-2 cells Concentrations 50 µg/ml Incubation Time 4 h Method BV-2 cells are maintained at 37℃ with 5% CO₂. To build OGD models, BV-2 cells are washed with PBS for 3 times, and cultured in glucose-free DMEN and placed in a hypoxia incubation chamber filled with the gas mixture for 4 h at 37℃. The culture medium is then replaced by the normal medium containing 4.5 g/L glucose and cells are cultured under normoxic conditions for reoxygenation for 24 h. BV-2 cells are treated with 50 µg/ml rmMFG-E8 or/and Colivelin TFA, respectively. The expression changes of proteins (p-STAT3 and SOCS3) are analyzed by Western blot assay.

Method
BV-2 cells are maintained at 37℃ with 5% CO₂.
