Description CK-666 is an inhibitor of the actin-related protein 2/3 (Arp2/3) complex with IC50 of 17 μM and 5 μM for BtArp2/3 complex and SpArp2/3 complex, respectively.

In vitro

In SKOV3 cells, CK-666 reduces actin polymerization around intracellular Listeria to background levels. Actin filament halos and comet tails reformed when CK-666 is removed after one hour of treatment.[1]

In vivo

CK-666 suppresses the enhanced memory phenotype of msi-1(lf) by inhibiting the Arp2/3 complex activity. In addition, inhibition of Arp2/3, involved in actin filament branching and neuronal morphogenesis, in the lateral amygdala (LA) with the specific inhibitor CK-666 during fear conditioning impaired long-term fear memory in rats.[2]
