Clear solution
5% DMSO 95% Corn oil
5.0mg/ml Taking the 1 mL working solution as an example, add 50 μL of 100 mg/ml clear DMSO stock solution to 950 μL of corn oil and mix evenly. The mixed solution should be used immediately for optimal results.
Clear solution
5%DMSO 40%PEG300 5%Tween80 50%ddH2O
2.5mg/ml Taking the 1 mL working solution as an example, add 50 μL of 50 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.
* <1 mg/ml means slightly soluble or insoluble.
* Please note that Selleck tests the solubility of all compounds in-house, and the actual solubility may differ slightly from published values. This is normal and is due to slight batch-to-batch variations.
* Room temperature shipping (Stability testing shows this product can be shipped without any cooling measures.)

Biological Activity

Description CDN1163 is a small molecular allosteric activator of the sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) that improves Ca2+ homeostasis.
Targets
SERCA [1]
In vitro

CDN1163 treatment significantly reduces the hepatic expression of genes involved in gluconeogenesis and lipogenesis, attenuates ER stress response and ER stress-induced apoptosis, and improves mitochondrial biogenesis, possibly through SERCA2-mediated activation of AMP-activated protein kinase pathway.[1]

In vivo

CDN1163 reduces blood glucose levels and improves metabolic parameters and gluconeogenic gene expression, reverses hepatic steatosis, inhibits ER stress and ER stress-induced apoptosis, and improves mitochondrial efficiency in ob/ob mice in vivo.

Protocol (from reference)

Cell Assay:

[1]

  • Cell lines

    HEK cells

  • Concentrations

    10 μm

  • Incubation Time

    2 h

  • Method

Cells are grown in 96-well plates (n = 6) and exposed to CDN1163 (10 μm) or vehicle (DMSO) for 2 h followed by the addition of 200 μm H2O2 compared with untreated controls. H2O2 activates the unfolded protein response and is an inducer of ER stress-promoted apoptosis. After 16 h of incubation, cell viability is measured using Luminescent Cell Viability Assay.
