Biological Activity

Description Capsaicin(Vanilloid) is an active component of chili peppers, which are plants belonging to the genus Capsicum.Capsaici is a TRPV1 agonist with an EC50 of 0.29 μM in HEK293 cells.Capsaicin is a mixture of Capsaicin and Dihydrocapsaicin.
In vitro

Capsaicin, a TRPV-1 agonist, dose-dependently inhibits the proliferation of the HCC cell lines PLC/PRF/7, HuH7 and HepG2, which is not effected by TRPV1 channel blockers. Combining capsaicin and sorafenib significantly enhances the suppression on cell proliferation, achieving a high-level synergistic effect and promoting HCC cell apoptosis through the enhancement of p-ERK and reduction of p-STAT3 signaling.[1]

In vivo

Combining capsaicin and sorafenib enhances inhibition of xenograft tumor growth in nude mice, with no apparent gross toxicity.[1]

  • Method

    Cells (5×103 per well) seeded in 96-well plates are exposed to capsaicin, sorafenib, or their combination for 72 h. Then, the medium is removed and the wells are washed with phosphate-buffered saline (PBS). The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay is performed by adding 20 μL of MTT to the cells for 4 h. Light absorbance is measured.
