Biological Activity

Description	Biocytin ((+)-Biocytin, Biotinyl-L-lysine) is a conjugate of D-biotin and L-lysine. Biocytin is an attractive neuronal tracer. It labels neurons fast over long distances, and it can be used in conjunction with calcium dyes to report on neuronal activity in retrogradely labeled live neurons.
In vitro	Biocytin labels type II cells via ion channels activated by [Ca²⁺]out reduction, probably "CALHM-like" channels, on the basolateral membrane.^[3]
In vivo	Biocytin confers stability to the molecules and increases their applicability as neuronal tract tracer.^[2]

Recording electrodes are attached to the basolateral membranes of single TBCs. In these voltage-clamp experiments, voltage-gated currents are amplified and filtered at 10 kHz using a voltage-clamp amplifier. Data are digitized using an A/D converter and are then stored using pCLAMP data acquisition and analysis software on a personal computer. The recording electrodes are filled with an intracellular CsCl solution using a pipette with a resistance of approximately 5 MΩ in physiological saline. To immunohistochemically identify cell types, biocytin (2 mg/ml) is injected through the recording electrodes.
