Biological Activity

Description Rhein (Monorhein, NSC 38629, Rheinic) is an anthraquinone compound enriched in the fresh rhizome of Rheum coreanum Nakai, showing anti-inflammation and antitumor activities. Rhein is an FTO (mRNA N6-methyladenine demethylase) inhibitor and also inhibits AlkB repair enzymes.
In vitro Rhein at 35 μM can effectively inhibit the synthesis of IL-1β on human osteoarthritis synovium, as well as the action of this cytokine at the cartilage level, by reducing the number of chondrocyte IL-1R and inhibiting IL-1β induced NO production in chondrocytes and cartilage. [1] [2] Rhein also induces apoptosis in human promyelocytic leukemia cells (HL-60), characterized by caspase activation, poly(ADP)ribose polymerase (PARP) cleavage, and DNA fragmentation. [3]
In vivo Rhein is also effective in ameliorating diabetic renal pathological changes and attenuating hyperlipidemia in db/db mice with diabetic nephropathy with decreased transforming growth factor-beta1 (TGF-beta1) and fibronectin immunohistochemistry expression in renal tissue and reduced cholesterol (Chol), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) and ApoE in plasma.
